Bacterial Flagella and Pili

Intermediate🦠 MicrobiologyTopic 19 of 54

Summary

Flagella propel cells while pili attach to surfaces and transfer DNA during conjugation.

In depth

The flagellum is a rotary motor powered by proton motive force that spins up to about 100 revolutions per second. Pili (fimbriae) mediate adherence; the sex pilus joins cells for conjugation.

Flagella arrangements: monotrichous, lophotrichous, peritrichous. Type IV pili allow twitching motility and DNA uptake.

Example: motility helps pathogens reach host cells.

The flagellar motor is a rotary engine with a stator and rotor using ions, and the filament is made of flagellin. Spirochaetes have periplasmic flagella. The flagellum is an example used to discuss the evolution of complex structures; it shares components with the type III secretion system. Type IV pili can extend and retract to pull cells along surfaces (twitching motility) and take up DNA.

Sex pili are used in conjugation. Adhesive pili are virulence factors, like the P pili of uropathogenic E. coli. Flagella are antigenic (H antigens) and are detected by the immune system through TLR5.

Example: Helicobacter pylori uses flagella to swim through stomach mucus to the epithelium.
