Ubiquitin-Proteasome System

Advanced🧫 Molecular BiologyTopic 43 of 52

Summary

Ubiquitin tagging marks proteins for destruction by the proteasome to control protein levels.

In depth

E1, E2 and E3 enzymes attach a chain of ubiquitin to a lysine of the target; a chain of four or more tags it for the 26S proteasome, which unfolds and cuts it into peptides. E3 ligases confer specificity, with hundreds in humans.

It controls cyclins, transcription factors and misfolded proteins.

Ubiquitin is a 76-amino-acid protein. Humans have two E1s, about 40 E2s and over 600 E3 ligases, the last of which provide specificity. The 26S proteasome has a 20S core cylinder with protease sites inside and 19S caps that recognise tags, unfold and feed the substrate with ATP. Aaron Ciechanover, Avram Hershko and Irwin Rose received the 2004 Nobel Prize.

The system removes misfolded proteins and controls short-lived regulators such as cyclins, p53 and IkB. Dysfunction contributes to neurodegenerative diseases, and proteasome inhibitors (bortezomib) treat multiple myeloma. PROTAC drugs harness it to degrade chosen targets.

Remember: ubiquitin tags can also signal non-degradation roles, depending on chain linkage.
